Longevity

NAD+

Half-life: 2h Subcutaneous or intramuscular injection; IV infusions are used clinically but cause flushing/nausea if pushed quickly.
Warnings. Rapid administration commonly causes flushing, chest pressure, and nausea - inject slowly.

Mechanism

Essential co-substrate for sirtuin (SIRT1-7) and PARP enzymes involved in DNA repair and mitochondrial biogenesis; central electron carrier in cellular respiration and ATP production.

Overview

NAD+ (nicotinamide adenine dinucleotide) is a critical coenzyme in cellular energy metabolism, DNA repair, and sirtuin activation. Levels decline substantially with age, and replenishment is studied for longevity, energy, and neuroprotection. Injectable NAD+ bypasses the limited oral bioavailability of direct NAD+ supplementation.
